Die Identifikation einer Mutation im Gen CARMIL2 bei Kindern mit angeborenem Immundefekt und gleichzeitiger Neigung zu Tumoren durch Münchner Forscher, deckt den Zusammenhang zwischen der Störung in den Immunzellen und der anti-tumoralen Immunität auf. Erkenntnisse der Wissenschaftler zur natürlichen Unterdrückung von Tumoren durch das Immunsystem bei immungeschwächten Patienten, eröffnen neue therapeutische Ansätze. Untersucht wurden im Rahmen der Internationalen Care-for-Rare Alliance, Kinder aus dem Nahen Osten und aus Südamerika, deren Körper mit Tumoren der glatten Muskelzellen befallen waren, die mit Epstein-Barr-Viren (EBV) in Verbindung gebracht werden konnten. Dabei konnten in den wichtigen Immunzellen, den sogenannten T-Lymphozyten, Störungen in einem Signalweg ausgemacht werden, die auf die Aktivierung des Oberflächenmoleküls CD28 zurückgeführt wird. Nun gilt dieser neue Signalweg als therapeutische Zielstruktur. Die Störung in den T-Lymphozyten verhindert die Ausreifung der T-Zellen und ihre Funktionsfähigkeit. Da das Immunsystem neben Infektionserregern auch eine wichtige Rolle bei der Verhinderung von Tumorentstehungen zuständig ist, spielen die beteiligten Signalwege bei betroffenen Patienten eine wichtige Rolle. Die Erkenntnisse der Arbeitsgruppe um Prof. Klein im Dr. von Haunerschen Kinderspital, wurden unter dem Titel „A human immunodeficiency syndrome caused by mutations in CARMIL2“ in der Zeitschrift „Nature Communications“ veröffentlicht. Beteiligt waren unter anderem auch Forscher des Max von Pettenkofer-Instituts, des Genzenzentrums, der TU München, des Helmholtz Zentrums und der Universidade Federal de Minas Gerais in Brasilien beteiligt. Für die Wissenschaftler ist die Erkenntnis des interdisziplinären Teams von entscheidender Bedeutung, wenn neue Therapien für Kinder mit lebensbedrohlichen Erkrankungen entwickelt werden sollen. Die Therapie sieht eine Steigerung der anti-tumoralen Immunität vor. Dies soll im Rahmen der Immun-Checkpoint-Blockade mittels hemmender Medikamente erreicht werden.
