Fusioniertes MET-Gen kann zu Gehirntumoren führen
Gefährliche und tödlich verlaufende Gehirntumore, die sogenannten Glioblastome, treten vermehrt bei erwachsenen Menschen auf. Doch auch Kinder sind nicht gegen das Enstehen dieser Krebsart gefeit. Bis zu 60 Kinder und Jugendliche werden jährlich von dieser Krankheit befallen. Die Behandlungen beschränken sich auf die Verzögerung des Tumorwachstums durch Strahlen- und Chemotherapie. Genetikforscher haben nun in einer Studie verschiedene Genveränderungen entdeckt, die zum Ausbruch der Krankheit führen können. Darunter das fusionierte MET-Gen, worüber sich neue Behandlungsmöglichkeiten ergeben.
Wissenschaftler des Max-Planck-Institus für molekulare Genetik und des Deutschen Krebsforschungszentrums, haben im Erbgut von Glioblastom-Tumoren einen unter vielen für den Ausbruch von aggressiven Hirntumoren verantwortlichen Gendefekt entschlüsselt. Bei zehn Prozent der betroffenen jungen Patienten konnte ein MET-fusioniertes Gendefekt nachgewiesen werden. In Deutschland erkranken jährlich etwa 60 Kinder an der nur schwer kaum behandelbaren Krebsart, den Glioblastomen. Durch die sogenannte Transkriptomanalyse konnte eine für die Tumorbildung verantwortliche Genmutation ausgemacht werden. In einer umfassenden Studie das von David Jones am Deutschen Krebsforschungszentrum in Heidelberg koordiniert wird, hat ein Team für molekulare Genetik um Marie-Laure Yaspo, am Max-Institut in Berlin, in Zusammenarbeit mit Forschern des Krebsforschungszentrums, eine Untersuchung zu Veränderungen der DNA und der Muster der Genaktivität von 42 an Glioblastomen erkrankten Kindern vorgenommen und ist dabei fündig geworden.
Dabei kam heraus, dass die Erkrankung durch viele verschiedene Veränderungen ausgelöst werden kann. Bei den genannten zehn Prozent konnte das fusionierte MET-Gen ausgemacht werden. Das für die Bildung eines Rezeptorproteins auf der Zelloberfläche verantwortliche Krebsgen MET, das auf Wachstumsfaktoren reagiert und das Zellwachstum kontrolliert, ist in diesem Gendefekt mit anderen DNA-Segmenten fusioniert. “Wir konnten ein MET-Fusionsgen bei mehreren Patienten nachweisen. Es könnte daher ein neuer Ansatzpunkt für eine individualisierte Behandlung von Glioblastom-Patienten sein“, erklärt Hans-Jörg Warnatz, einer der Erstautoren der Studie. Er gehört zu jenen Forschern, die die Genaktivität innerhalb der Tumore untersucht und die Genfusion gefunden haben. Bei einer Überfunktion des MET-Gens bildet eine Zelle zu viel dieser Rezeptoren, wodurch das Wachstum von Krebs angeregt wird. Durch die Fusion mit weiteren Abschnitten der DNA geht die normalerweise strenge Aktivitätskontrolle verloren. MET-Fusionsgene sind nach einer Gruppe Wissenschaftler um Stefan Pfister und Peter Lichter vom Deutschen Krebsforschungszentrum in Heidelberg erwiesenermaßen für die Entstehung von Glioblastomen verantwortlich. Nach der Transfusion von MET-Fusionsgenen in die Hirnzellen von Mäusen, entwickelten diese die gleichen Tumore wie menschliche Patienten. Der Einsatz eines MET Hemmstoffs, welches bereits bei anderen Krebsformen in Gebrauch ist, kann bei Hirntumoren vorerst nicht angewendet werden. Vereinzelte Resistenzen bei Patienten und die Gefahr weiterer bösartiger Mutationen neben den MET-Fusionsgenen erfordern weitere Untersuchungen. Yaspo, Leiterin der Transkriptomanalysen in Berlin machte deutlich, dass die molekulare Analyse individueller Tumore neben der Mutationen auch Änderungen im Transkriptom identifiziert und somit zum besseren Verständnis der einzelnen Krankheitsverläufe beiträgt.
