Der von im Boden lebenden Mikroorganismen gebildete antibiotische Naturstoff Carolacton hat sich nach neuesten wissenschaftlichen Untersuchungen als zweischneidiges Schwert erwiesen. Aufgrund seiner inhibitorischen und bindenden Wirkung auf ein wichtiges Schlüsselenzym, das an der Bildung von Folsäure im C1-Stoffwechsel beteiligt ist, ist es zwar als Antibiotikum kaum einsetzbar, könnte aber bei optimierter Nutzung Krebszellen in ihrem Wachstum ausbremsen. Das haben die Forscher Chengzhang Fu und Rolf Müller vom Helmholtz- Institut für Pharmazeutische Forschung Saarland (HIPS) und vom Helmholtz-Zentrum für Infektionsforschung (HZI) in ihren Untersuchungen mit Modellorganismen herausgefunden. Die Ergebnisse der in-Vitro Forschungen wurden im Fachjournal Nature Communications veröffentlicht. Dadurch ist Carolacton nun in den Focus der therapeutischen Behandlung von Krebs gerückt. Carolacton ist in der Lage wichtige Prozesse in Bakterien zu unterbinden. Als Wachstumsinhibitor von Streptococcus pneumonieae (lebensbedrohlichen Mikroorganismen), hemmt es jedoch auch das eigentlich wichtige Schlüsselenzym FolD, das Krebszellen nach bisherigen Erkenntnissen mehr benötigen als gesunde Zellen. Carolacton kann das Enzym FolD binden und ausschalten. Dieses Enzym ist jedoch unverzichtbar, da es an der in den meisten Lebewesen vorkommenden Bildung von Folsäure im C1 Stoffwechsel beteiligt ist. Über diesen Prozess werden Nukleinsäuren, Aminosäuren und Provitamine hergestellt. Diese Tatsache erschwert den Einsatz von Carolacton als Antibiotikum. Die Forscher konnten das bislang unbekannte molekulare Ziel des Wirkstoffs in den Modellorganismen Escherichia coli und Streptococcus pneumoniae ausmachen. Für den Umgang mit Krebszellen eröffnet diese Erkenntnis neue Möglichkeiten in Forschung und therapeutischen Ansätzen. „Wir konnten in ersten Experimenten hemmende Effekte bei verschiedenen getesteten Krebszelllinien zeigen“, konkretisiert Müller im Deutschen Ärzteblatt.
