Am Max-Planck-Institut für terrestrische Mikrobiologie in Marburg ist es dem Wissenschaftler Tobias Erb und seinen Mitarbeitern gelungen, CO2 aus der Luft mittels neu entwickeltem synthetischen Stoffwechsel in eine organische Substanz umzuwandeln,- und das effizienter als es durch die Fotosynthese den Pflanzen gelingt. In einem Interview mit dem MPI sprach der Wissenschaftler von der Tragweite der Technologie für den Klimaschutz und den Perspektiven der synthetischen Biologie in der Zukunft.
Im Vordergrund stehe die Aufgabe, grundlegende biologisch-chemische Prinzipien zu verstehen, um nachzuvollziehen wie gasförmiges CO2 in organische Moleküle umgewandelt werden kann. Das Ziel der Wissenschaftler, atmosphärisches CO2 mit biologischen Methoden als Kohlenstoffquelle der Zukunft zu erschließen birgt positive Nebeneffekte. Doch Erb nimmt dazu klare Haltung. Der positive CO2-neutrale Prozess zu dem man dabei komme, oder gar die Reduktion von CO2 in der Atmosphäre, seien lediglich tolle Nebeneffekte. Die Motivation der Forschung entspringe nicht primär einer Idee den Klimawandel zu stoppen. Der Wissenschaftler glaubt, dass sogenannte Designer-Stoffwechselwege wie der geringere Energieverbrauch pro umgesetztem CO2-Molekül oder die schnellere Bindung von CO2 aus der Luft interessante Ansätze zur Schaffung CO2 basierter Biotechnologien seien. Noch steht die Technologie am Anfang. Es sei das erste Mal, dass die Umwandlung von CO2 in organische Substanzen im Reagenzglas imitiert werden konnte. Dies unterscheide sich vollkommen von der Herausforderung metabolische Organe in lebende Organismen zu transplantieren.
Was die erfolgreiche Transplantation angeht, spricht der Wissenschaftler von einem großen Reaktionsunterschied zwischen dem Stoffwechsel der synthetisch entstandenen organischen Substanz mit 17 Reaktionen und dem der Zelle eines lebenden Organismus mit 3000 gleichzeitig verschiedenen Reaktionen. Dementsprechend brauche die Transplantation noch viel Zeit. Trotz der Berechnungen die eine effizientere Umwandlung vermuten lassen als die Stoffwechselwege der Pflanzen, müsse diese Theorie im Experiment zunächst nachgewiesen werden.
Nach dem der rein technische Entwurf des Stoffwechsels fertiggestellt war, folgten zwei Jahre der experimentellen Umsetzung des Zyklus. Dann ging es daran, alle biologischen Einzelteile des Zyklus, die Enzyme zu finden und zusammenzustellen. Hierzu kamen die Forscher nicht herum, aus mehr als 50 Millionen bekannten Genen und 40.000 Enzymen potenzielle Enzyme ausfindig zu machen. Die Arbeit als Biologe mit synthetischen Ansätzen bezeichnete Erb als immer noch ungewöhnlich. Man sei eher gewohnt biologische Systeme auseinanderzunehmen und zu analysieren. Da den Wissenschaftlern nur wenige Regeln für das Design synthetischer Systeme bekannt seien, mussten sie sich langsam vortasten. Zunächst, so Erb, müsse herausgefunden werden, welche Enzyme zueinander passen und worauf beim Zusammenbau eines komplexen biologischen Systems geachtet werden muss.
Da die Forscher mit einberechnen müssen, dass die in den Mitochondrien, Ribosomen und anderen Organellen stattfindenden zellulären Prozesse natürlich bedingt voneinander getrennt ablaufen, ist eine zusätzliche Herausforderung gegeben. Man müsse noch herausfinden, wann genau biochemische Reaktionen voneinander getrennt werden können. Erb distanziert sich von wahnwitzigen Ideen lebende Zellen zu schaffen. Man wolle in keinem Fall Gott spielen. Viel näher lägen dann schon die Versuche einzelne Lebensprozesse umzuprogrammieren, wie es gerade im MaxSynBio Netzwerk der Max-Planck-Gesellschaft vorgenommen werde. Der Nachbau biologischer Prozesse lasse auf neue Ideen kommen, da hierbei sehr viele Enzyme bei der Arbeit gesehen werden können. Erb betont in diesem Zusammenhang auch, dass biologische Systeme effizienter und unter milderen Bedingungen arbeiten. Doch wo führen Forschungen die zur Ergründung biologisch-chemischer Systeme beitragen und wo genau könnte die synthetische Biologie ihren Einsatz finden? Erb lässt verstehen, dass das vorhandene Potenzial diesbezüglich kaum einzuschätzen ist und hält den Vergleich mit der sich ab dem 19. Jahrhundert von einer analytischen zu einer synthetischen Disziplin entwickelten Chemie am geeignetsten. Er ist zuversichtlich, dass der synthetischen Biologie ein ähnlicher Weg bevorsteht.
